PSX-B-12 Effect of Locust Bean gum on Gene Expression in cow Blood

Abstract Locust bean gum (LBG) is a galactomannan extracted from the seeds for use as food thickener and stabilizer claimed to have applications in biopharmaceutical field. The possible immunomodulatory effects of LBG been studied cell lines. objective this study was evaluate effect on expression genes associated with innate adaptive immune responses cow blood. Blood collected (n=3) clinically healthy Holstein-Friesian cows North Carolina A&T State University Dairy Unit. Whole blood (1 mL) three treated individually 10 ng/mL (Sigma-Aldrich St. Louis, MO) or maintained PBS, incubated at 37ºC 30 minutes. Total RNA extracted, reverse transcribed, real-time PCR carried out using RT2 Profiler human Innate & Adaptive Immune Responses Array containing 84 genes, recommended by manufacturer (Qiagen). Livak method used calculate fold change (FC >2 considered significant). analysis showed that 40 were affected treatment LBG. Among 4 upregulated, 17 downregulated. highest up-regulated down-regulated following exposure Metaxin 1(MX1) Recombination activation gene 1 (RAG1) respectively.
